This method is mainly used to study the interactions between biomolecules, such as protein-protein, protein-DNA, protein-small molecule drugs, etc. The basic principle of MST is that when a small temperature gradient is applied to a solution containing biomolecules, these molecules will move towards a colder or hotter direction according to their size, charge, and the properties of the solvation shell. This phenomenon is called thermophoresis. MST uses this principle to infer the interactions between molecules by measuring the movement of target molecules (usually fluorescently labeled) in a microchannel when a temperature gradient is applied.
