In the cost structure of ATP assay kits, recombinant luciferase accounts for the largest share, reaching 50%–60% of the total cost. Although its expression and purification processes (such as Ni chelation chromatography) are mature, significant batch-to-batch variations in enzyme activity require additional quality control, driving up the unit cost. D-luciferin substrate follows closely, accounting for approximately 15%–20%; high-purity (≥99%) potassium salt form still costs several thousand yuan per gram and is sensitive to humidity, requiring nitrogen-filled aluminum foil packaging, further increasing raw material costs. ATP standards, buffer salts, swabs, or microplates and other consumables account for 15%–20% combined, with ATP standards requiring extremely high purity and stability, costing over a thousand yuan even in small quantities. The remaining 5%–10% covers cold chain transportation, dry and light-proof packaging, and quality control (including luminescence analyzer calibration and batch parallel testing). The overall structure is rigid, and economies of scale have limited impact on cost reduction.
