Acridine Orange Staining Solution is a fluorescent dye, which has membrane permeability and can stain nuclear DNA and RNA through cell membrane. Its excitation wavelength is 488nm and absorption wavelength is 515nm. It is different from the amount of DNA and RNA binding in cells. The complex can emit different colors of fluorescence, red fluorescence is AO-DNA(F>600nm), green fluorescence is AO-RNA or single-stranded DNA(F>530nm). Therefore, under fluorescence microscope, acridine orange can penetrate normal cell membrane and make the cell nucleus appear green or yellow-green uniform fluorescence. In apoptotic cells, the chromatin shrinks or breaks into fragments of different sizes, forming apoptotic bodies. Acridine orange causes it to be stained with dense yellow-green fluorescence, or yellow-green debris particles; The yellow fluorescence of necrotic cells decreased or even disappeared. Acridine orange AO is often combined with ethidine EB bromide for double staining. Because EB only stains dead cells to produce orange fluorescence, normal cells, apoptotic cells and necrotic cells can be distinguished.
